Being a bit of a car camper I'm not used to lugging everything in for miles so I need to be organised, especially taking all those tinnies in.

So the question is: is it feasible to make a couple of journeys from car to campsite at V Fest or is there a sort of one-way barrier that you can't get back to the car from once inside V Fest?

Any tips about where best to camp also gratefully received.......the info on the website seems curiously silent on these questions although anecdotally I've heard "don't pitch by the walkways"

You can walk to and fro your car, it can be a long way though. Make as many trips as you need. Camp where you like, go find a nice looking space, don't camp near walkways is good advice though.
